Large Genomic Library Can Reveal the Drug Resistance Gene of Deadly Fungus

The genomic library for Candida glabrata is one of the three major genomic libraries of Candida glabrata world. Through the genomic library construction and analysis of Candida glabrata mutant, the researchers found 28 genes that made Candida glabrata had resistance to common drugs.

Infectious diseases caused by fungi, viruses, bacteria and parasites are main reasons causing human deaths, and about 1,500,000 deaths are due to infection of harmful fungi every year. In this study, the researchers were searching for resistance gene for Candida glabrata. Through the construction for Candida glabrata mutant library, they carried out the genetic analysis, and finally found 28 genes related with Candida glabrata resistance, especially the gene resistant to drug caspofungin.

At the same time, researchers also identified the new cell pressure sensors and transmitters in Candida glabrata. Removing the characteristics of Candida glabrata would make it susceptible to antifungal therapy drugs, including caspofungin. Removing the virulence factor from the patient's body with the genetically method, will reduce the pathogenicity of Candida albicans and also increase its sensitivity to the drug.

Currently the world has spent more than 80 million euros in the research and development of anti fungal drugs. Infections triggered by Candida glabrata often require more expensive drugs, such as caspofungin. Caspofungin can block the synthesis of the cell wall with rich carbohydrates of fungal cells. Due to resistance gene of Candida glabrata, it produces a strong resistance to the traditional drugs, like caspofungin.
